Recently, we reported that titanium dioxide (TiO2) materials activated endothelial cells via Kruppel-like factor (KLF)-mediated nitric oxide (NO) dysfunction, but the roles of physical properties of materials are not clear. In this study, we prepared nanobelts from P25 particles and compared their adverse effects to human umbilical vein endothelial cells (HUVECs). TiO2 nanobelts had belt-like morphology but comparable surface areas as P25 particles. When applied to HUVECs, P25 particles or nanobelts did not induce cytotoxicity, although nanobelts were much more effective to increase intracellular Ti element concentrations compared the same amounts of P25 particles. Only nanobelts significantly induced THP-1 adhesion onto HUVECs. Consistently, nanobelts were more significant to induce the expression of intracellular adhesion molecule-1 (ICAM1) and the release of soluble ICAM-1 (sICAM-1), indicating that nanobelts were more potent to induce endothelial activation in vitro. As the mechanisms for endothelial activation, both P25 and nanobelts reduced the generation of intracellular NO as well as the expression of NO regulators KLF2 and KLF4. Combined, the results from this study indicated that the different morphologies of P25 particles and nanobelts only changed their internalization into HUVECs but showed minimal impact on KLF-mediated NO signaling pathways. 相似文献

[摘要] 目的 探讨ICU患者发生产超广谱β-内酰胺酶(extended spectrum beta-lactamases, ESBLs)革兰阴性杆菌感染的危险因素,并构建相关预测模型。方法 选取2017年5月—2021年4月我院ICU发生大肠埃希菌或肺炎克雷伯菌感染的189例患者作为研究对象,收集患者的临床资料,使用单因素分析、LASSO回归和多因素Logistic回归分析ICU患者30 d内发生产ESBLs革兰阴性杆菌感染的危险因素,并据此建立列线图预测模型。结果 急性生理与慢性健康评分≥16分、留置尿管时长≥7 d、抑酸剂使用时长≥3 d、第三代头孢菌素使用时长≥3 d、抗菌药物联用时长≥3 d和ICU住院时间≥15 d是ICU患者30 d内发生产ESBLs革兰阴性杆菌感染的危险因素(P均<0.05)。依此建立预测ICU患者30 d内发生产ESBLs革兰阴性杆菌感染的列线图风险模型,模型验证结果显示C-index为0.795,校正曲线趋近于理想曲线,AUC为0.807(95%CI:0.775~0.839),在2%~81%预测范围内,列线图净获益。结论 ICU患者30 d内发生产ESBLs革兰阴性杆菌感染的危险因素包括APACHEⅡ评分≥16分、留置尿管时长≥7 d、抑酸剂使用时长≥3 d、第三代头孢菌素使用时长≥3 d、抗菌药物联用时长≥3 d和ICU住院时间≥15 d,据此构建的列线图模型能有效预测ICU患者30 d内发生产ESBLs革兰阴性杆菌感染的风险概率,具有一定的临床价值。 相似文献

目的 分析南京市某国际高中学生艾滋病知识知晓率及其影响因素,为制定高中生艾滋病健康教育策略提供依据。方法 对南京市某国际高中所有学生开展纸质问卷调查,统计分析艾滋病知识知晓率及其影响因素。 共纳入171名高中生为调查对象,艾滋病和梅毒的知识知晓率分别为56.1%和46.8%;有过性行为的占14.6%,其艾滋病知识知晓率最高为32.0%,(χ2=10.799,P=0.005);有吸毒史的学生占9.3%,其艾滋病知识知晓率最高为31.2%,(χ2=21.062,P<0.001);梅毒知识知晓者与不知晓者的艾滋病知识知晓率差异具有统计学意义(χ2=21.716,P<0.001)。 该国际高中学生性病和艾滋病知识知晓率较低,对高中生开展毒品危害教育和性病艾滋病防治应作为健康教育工作重点。 相似文献

In 2019, the scientists who discovered how cells sense and adapt to oxygen availability were awarded the Nobel Prize. This elegant sensing pathway is conserved throughout evolution, and it underpins the physiology and pathology that we, as clinicians in anaesthesia and critical care, encounter on a daily basis. The purpose of this review is to bring hypoxia-inducible factor, and the oxygen-sensing pathway as a whole, to the wider clinical community. We describe how this unifying mechanism was discovered, and how it orchestrates diverse changes such as erythropoiesis, ventilatory acclimatisation, pulmonary vascular remodelling and altered metabolism. We explore the lessons learnt from genetic disorders of oxygen sensing, and the wider implications in evolution of all animal species, including our own. Finally, we explain how this pathway is relevant to our clinical practice, and how it is being manipulated in new treatments for conditions such as cancer, anaemia and pulmonary hypertension. 相似文献

目的:探讨基于肿瘤浸润深度及大小的TTS评分系统对可切除胃癌患者预后判断的价值。方法:选择行根治性切除的234例胃癌患者为研究对象。根据肿瘤浸润深度(T分期)和肿瘤大小构建TTS评分系统,分为TTS 0级、1级、2级,代表肿瘤侵袭性逐渐升高。通过Cox多因素模型分析TTS评分系统作为预后判断工具的可行性。结果:T1-T4期患者肿瘤的平均大小分别为(3.3±2.7)cm、(4.1±3.1)cm、(6.6±3.1)cm、(9.4±4.9)cm。单因素分析显示,肿瘤浸润深度与肿瘤大小、淋巴结转移、TNM分期、淋巴浸润和血管浸润显著相关(P<0.01)。ROC曲线显示,45 mm为肿瘤大小的最佳界值,可有效区分患者是否存在淋巴结转移,曲线下面积(area under curve,AUC)为0.762。综合肿瘤大小的临界值和肿瘤浸润深度,构建TTS评分系统,TTS 0级、TTS 1级和TTS 2级的患者5年生存率分别为95.6%、83.3%和70.2%,两两比较后发现,不同TTS状态患者的生存率之间均存在显著差异(P<0.01)。Cox多因素分析发现,TTS评分是影响患者预后的独立性危险因素(P<0.05)。结论:本研究根据肿瘤浸润深度和大小构建了TTS评分系统,并证明了TTS评分与胃癌患者的预后密切相关。 相似文献

PurposeTo quantify joint degeneration and the clinical outcome after curettage and cementation in subchondral giant cell tumors of the bone (GCTB) at the knee.MethodsWe conducted a retrospective analysis of 14 consecutive patients (seven female, seven male) with a mean age of 34 years (range 19–51) who underwent curettage and subchondral cementation for a biopsy-confirmed GCTB at the distal femur or the proximal tibia between August 2001 and August 2017, with a mean follow-up period of 54.6 months (range 16.1–156 months). The Whole-Organ Magnetic Resonance Imaging Score (WORMS), Kellgren-Lawrence (KL) classification, and Musculo-Skeletal Tumor Society (MSTS) score were assessed.ResultsRadiological degeneration progressed from preoperative to the latest follow-up, with a median WORMS from 2.0 to 4.0 (p = 0.006); meanwhile, the median KL score remained at 0 (p = 0.102). Progressive degeneration (WORMS) tended to be associated with the proximity of the tumor to the articular cartilage (mean 1.57 mm; range 0–12 mm) (p = 0.085). The most common degenerative findings were cartilage lesions (n = 11), synovitis (n = 5), and osteophytes (n = 4). Mean MSTS score increased from 23.1 (preoperatively) to 28.3 at the latest follow-up (p < 0.01).Seven patients (50%) were treated for a local recurrence, with six revision surgeries performed. Removal of the cement spacer and filling of the cavity with a cancellous autograft was performed in seven patients. Conversion to a total knee arthroplasty was performed in one patient for local tumor control.ConclusionsCementation following the curettage of GCTB around the knee is associated with slight degeneration at medium-term follow-up and leads to a significant reduction in pain. Intratumor heterogeneity is a main cause of the dismal prognosis of glioblastoma (GBM). Yet, there remains a lack of a uniform assessment of the degree of heterogeneity. With a multiscale approach, we addressed the hypothesis that intratumor heterogeneity exists on different levels comprising traditional regional analyses, but also innovative methods including computer-assisted analysis of tumor morphology combined with epigenomic data. With this aim, 157 biopsies of 37 patients with therapy-naive IDH-wildtype GBM were analyzed regarding the intratumor variance of protein expression of glial marker GFAP, microglia marker Iba1 and proliferation marker Mib1. Hematoxylin and eosin stained slides were evaluated for tumor vascularization. For the estimation of pixel intensity and nuclear profiling, automated analysis was used. Additionally, DNA methylation profiling was conducted separately for the single biopsies. Scoring systems were established to integrate several parameters into one score for the four examined modalities of heterogeneity (regional, cellular, pixel-level and epigenomic). As a result, we could show that heterogeneity was detected in all four modalities. Furthermore, for the regional, cellular and epigenomic level, we confirmed the results of earlier studies stating that a higher degree of heterogeneity is associated with poorer overall survival. To integrate all modalities into one score, we designed a predictor of longer survival, which showed a highly significant separation regarding the OS. In conclusion, multiscale intratumor heterogeneity exists in glioblastoma and its degree has an impact on overall survival. In future studies, the implementation of a broadly feasible heterogeneity index should be considered. 相似文献
